Cell Line Development (CLD) & Discovery

Cell Line Development is the foundation of biopharmaceutical production. Establishing monoclonality is not just a biological requirement; it is a regulatory mandate. The data-driven decisions made during these early stages define the safety, consistency, and scalability of your future manufacturing process.

We provide an integrated workflow that ensures regulatory confidence and accelerates the transition from single cell to suspension culture.

  • Automated Single-Cell Deposition: Using the Seed Biosciences Dispencell S4, we enable high-efficiency single-cell seeding across 96-well plates in less than 3 minutes. This provides the first of two critical pieces of evidence required for FDA/EMA regulatory proof of clonality.
  • High-Throughput Imaging: We integrate SYNENTEC systems (NYONE and CELLAVISTA) to provide the definitive image-based proof of monoclonality (2/2). These platforms also automate confluence measurements and image-based assays, whether as standalone units or within integrated automation tracks.
  • Rapid Clone Assessment: Leadgene Biosolutions enables researchers to perform rapid assessments in suspension culture. With pH and dissolved oxygen (DO) measurement in microwell plate formats, you gain critical insight into clone performance earlier in the timeline.
  • Early-Stage Expansion: Thomson Optimum Growth® plates and flasks are essential here. Transitioning cells from plates to shake flasks is the first "scale-up" hurdle; using Thomson technology at this stage ensures consistent aeration and growth characteristics as you move toward Upstream Processing.

Upstream Processing (USP)

Upstream success relies on maintaining cell health and productivity while navigating the complexities of scale-up. BPES provides the tools to transition seamlessly from CLD through process development to pilot scale and beyond.

Seamless Scale-Up and Monitoring

  • Optimised Cell Expansion: Thomson Optimum Growth® flasks (up to 7L) utilise a unique baffled design and wide neck to increase aeration, closely mimicking the $k_La$ values found in bioreactor environments. This maximises incubator space while ensuring your cells are "bioreactor-ready."
  • Precision Bioreactors: We provide Solaris Biotech solutions ranging from 200mL benchtop units to 50,000L manufacturing scales. These systems offer fully customisable S/I series options and cGMP-compliant software for rigorous process control.
  • Real-Time Biomass Insight: To eliminate the risks of manual sampling, we utilise ABER FUTURA capacitance-based sensors. This provides online monitoring of viable cell density (VCD), allowing you to identify deviations immediately and understand biomass trends without breaking the sterile boundary.
  • Custom Sterile Fluid Transfer: We understand that your process is unique. Through HPConnex, we design custom single-use assemblies (in partnership with High Purity New England) tailored to your specific sampling and transfer requirements.

Primary Recovery & Harvest

  • Low-Shear Harvest: We utilise Carr Biosystems single-use tubular bowl centrifugation. This technology provides a scalable, low-shear solution (bench to 2,000L) that significantly reduces LDH release compared to traditional methods. By delivering a consistent starting point for downstream operations, it reduces the burden on depth filters and slashes the required filter surface area.

 

 

Downstream Processing (DSP)

Protecting the structural integrity of sensitive biological molecules is the final critical hurdle. After weeks of nurturing a cell culture, the focus shifts to purifying the "golden product" while maintaining high yields.

Controlled Liquid Conveyance

  • Low-Shear Fluid Handling: Quattroflow® quaternary diaphragm pumps (available in single-use or stainless steel) provide the gentle, low-pulsation handling necessary to "protect every drop" during sensitive filtration and chromatography steps. With single-use and multi-use options and flow rates from 1mL/min to 1600L/h, they support every scale.

High-Precision Dosing: When paired with PSG Biotech flow measurement solutions, manufacturers achieve a rigorously controlled, scalable process that ensures unit operations perform predictably every time.
